Study Finds No Causal Link Between Pregnancy Inflammation and Child Neurodevelopmental Outcomes

Research Overview

A recent study investigated whether inflammation during pregnancy has a direct impact on neurodevelopmental outcomes in children. Using genetic data from the Norwegian Mother, Father and Child Cohort Study (MoBa), researchers explored potential causal relationships between maternal inflammation markers and offspring neurodevelopmental traits, including attention-deficit/hyperactivity disorder (ADHD) and autism diagnoses.

Study Methods

The research team employed advanced genetic approaches, specifically trio polygenic score (PGS) and intergenerational Mendelian randomization (MR) analyses. These methods utilized genetic information from mothers, fathers, and children simultaneously to isolate effects attributable to maternal inflammation during pregnancy, while accounting for familial genetic confounding.

Inflammation was measured through genetic proxies for three biomarkers: C-reactive protein (CRP), interleukin-6 (IL-6), and glycoprotein acetyls (GlycA). Neurodevelopmental outcomes were assessed based on mother-reported traits from ages 3 to 8 and lifetime clinical diagnoses of ADHD and autism.

Key Findings

Validation of genetic instruments showed strong predictive power only for CRP. Analyses involving 41,531 complete parent-child trios found no evidence that maternal genetic predisposition to higher CRP levels during pregnancy indirectly affected offspring neurodevelopmental outcomes. Similarly, Mendelian randomization analyses did not support a causal effect of maternal inflammation, as proxied by CRP, on children’s neurodevelopment.

Context and Implications

Previous observational studies have suggested links between pregnancy inflammation and child neurodevelopmental conditions. However, this genetically informed study indicates that such associations may be influenced by shared genetic factors rather than a direct causal effect of maternal inflammation during pregnancy.

These findings suggest that interventions targeting inflammation in pregnancy might not directly influence neurodevelopmental outcomes in offspring. Nevertheless, the authors note that other factors, such as selective participation in the cohort, could influence results, and further research is warranted.

Limitations and Considerations

The study focused on genetic proxies for inflammation markers, which may not capture all aspects of inflammatory processes during pregnancy. Additionally, the cohort’s characteristics and participation patterns might limit the generalizability of findings. As with all genetic studies, results indicate associations consistent with or against causality but do not provide definitive proof.
