Early-Life Phthalate Exposure May Influence Autism Traits Differently in Boys and Girls: Systematic Review

New Review Explores Phthalates and Autism Spectrum Traits

A recent systematic review has examined the potential link between early-life exposure to phthalates—chemicals commonly found in plastics and personal care products—and autism spectrum disorder (ASD) traits, with a focus on differences between sexes. The review analyzed 18 studies assessing prenatal and early childhood exposure to phthalates and their association with autistic behaviors.

Key Findings on Sex-Specific Vulnerability

The review found that prenatal exposure to certain phthalates, particularly during early to mid-pregnancy, was associated with subtle increases in autistic traits in children. Some evidence suggested that boys might be more susceptible to these effects, especially concerning metabolites of phthalates such as monobutyl phthalate (MBP), monoethyl phthalate (MEP), and di(2-ethylhexyl) phthalate (DEHP).

Assessment Methods and Study Characteristics

Phthalate exposure was primarily measured using urinary biomarkers collected from mothers or children, with some studies also using blood samples. The studies included children from infancy up to 15 years old, with sample sizes ranging from 77 to over 3,000 participants. Autism-related outcomes were evaluated using validated tools like the Social Responsiveness Scale (SRS) and the Autism Diagnostic Observation Schedule (ADOS).

Context and Importance of Findings

ASD is a neurodevelopmental condition marked by challenges in social communication and repetitive behaviors, and it is diagnosed approximately three times more often in boys than girls. While the biological reasons for this sex difference remain unclear, environmental factors such as exposure to endocrine-disrupting chemicals during critical developmental windows are being investigated as potential contributors.

Implications for Families and Caregivers

These findings highlight the importance of understanding how environmental exposures during pregnancy and early childhood might influence neurodevelopment differently in boys and girls. Although the associations observed are subtle and do not establish causation, they underscore the need for cautious use of products containing phthalates, especially during pregnancy.

Limitations and Need for Further Research

The review authors emphasize that the current evidence is limited and varies across studies in terms of exposure assessment and outcome measurement. More research is needed to confirm these findings and to better understand the mechanisms underlying sex-specific vulnerability to phthalates and ASD traits.
